Azaspiracid accumulation, detoxification and biotransformation in blue mussels (Mytilus edulis) experimentally fed Azadinium spinosum ArchiMer

Azadinium spinosum (Elbrächter and Tillmann), a small marine dinoflagellate, has been recently described as a de novo producer of azaspiracid-1 and -2 (AZA1 and -2) diarrhoeic toxins. A culture of A. spinosum was established in our laboratory and optimised for pilot-scale production of this organism, to evaluate and understand AZA1 and -2 accumulation and biotransformation in blue mussels (Mytilus edulis) fed with A. spinosum. Adult mussels were continuously exposed to A. spinosum over 1 week in 160 L cylindrical conical tanks. A bloom of the toxic dinoflagellate Alexandrium tamarense was observed for the first time in Thau iagoon between November and December 1998. Maximum cell concentrations detected in shellfish farming areas (85 000 Cells/1) were high enough to make mussels and clams unsuitable for human consumption (max. 850 ug. STX eq.100 g'1) while toxic levels in oysters did not exceed, or even reach, the international public health threshold established for PSP toxins. Blooms of Dinophysis in French coastal waters are implicated in most bans on marketing commercial bivalves. However, the relation between Dinophysis cell density and shellfish toxicity is not always consistent. Discrepancies may be due to the simple fact that it is nearly impossible to compare an integral over a few days (shellfish toxin content) and water samples. Furthermore, it seems that cells may have a variable specific toxicity. Development of cost effective tools for risk management and traceability systems for marine biotoxins in seafood. ArchiMer

The objective of the study was to pinpoint a possible effect of the food on the decontamination speed up of mussels containing DTX toxins. Studying the possible impact of food on the detoxification rate consists in i) following the toxin content over time for both feeding conditions and ii) checking the physiological state of the animals. For this purpose, an experimental protocol has been designed to describe the kinetics and to compare the detoxification rate in fed and non fed toxic mussels. Following exceptional outbreak of toxic dinoflagellate species in Vilaine Bay (summer 1983) a survey was carried out: variations in phytoplankton abundance and distribution in water and in mussel guts were followed. In the same time toxicity studies using mouse-test were performed with contaminated shellfishes, and mouse-test calibration was attempted. In spite of sampling and analysis conditions could be open to criticism, results obviously showed relation between shellfishes toxicity and occurrence of responsible species: Dinophysis acuminata.

Etude de la décontamination de moules toxiques (toxines diarrhéiques) en laboratoire et en milieu naturel ArchiMer

Two studies of depuration, one in laboratory conditions and the other one in an oyster's pond were performed using 2 sets of contaminated mussels with diarrhetic toxins at a high and a low level (3 and 1 M.U.). Depuration kinetics were followed both by the adult mouse test and okadaïc acid analysis. Results are - in the pond, high toxic mussels need about 20 days to become eatable (toxicity level : 0.5 M.U.g-1 digestive glands), -this time decreases when the initial toxicity is low but it increases when mussels are underfed, - a good correlation is observed between the total toxicity and okadaïc acid concentration.

The use of rotenone as a means of predators eradication is necessary for the success of shrimps culture. Because these compound is a fish toxicant, some precautions have to be tacken in order to avoid the contamination of marine environment by the residual waters ; so the dissipation and the toxicity have been studied together in laboratory and in field assays. It appears that a delay of 15 days should be respected : this time is required for a total disappearing of the residual toxicity.

A cytodetection test of diarrhoeic phycotoxins was developped by both IFREMER and Nantes University. Its principle is based on cell morphological changes of KB strain, induced by inhibition of protein-phosphatases involved in cytoskeleton building. This test has been shown sensitive, fast and specific for protein phosphatase inhibitors like diarrhoeic toxins (specialy okadaic acid in France). Industrial scale detoxification of phycotoxin-contaminated shellfish : myth or reality ? ArchiMer

As early as 1999, the EUROHAB initiative reported an urgent need for research on phycotoxin accumulation, detoxification and biotransformation rates in exploited shellfish species. It also strongly recommended a second objective of developing commercial systems for the cleansing of shellfish once contaminated with toxin. As a result, EU Framework Programs and regional programs funded a series of R & D projects aiming to consolidate our understanding of contamination/detoxification pathways, together with the development of industrial scale detoxification processes and mechanisms. ISSR as new markers for genetic characterization and evaluation of relationships among phytoplankton ArchiMer

In order to increase the molecular tools and markers needed for the identification of phytoplankton species, the inter simple sequence repeat ( ISSR) fingerprinting was adapted to micro-algae and its use in genetic analysis was demonstrated. Twelve strains, 6 Alexandrium, 4 Pseudo-nitzschia, 1 Skeletonema and 1 Tetraselmis were analysed for the first time with ISSR amplifications. The patterns were highly polymorphic and very reproducible. The 6 primers gave 223 polymorphic markers that clearly and easily distinguished all 12 strains ( mainly toxic ones) and gave 187 polymorphic markers among the Alexandrium and the Pseudo-nitzschia species. Mise au point d'un test microcrustacé marin ArchiMer

Because of the low number of currently available toxicity tests for marine environment, the authors describe an experimental procedure using a marine copepod : Tigriopus brevicornis. This method can be used to quantify the 50 % inhibition of larval production or growth. The variability in controls larval production reaches about 9 %. The toxicity of two contaminants : tributyltin (TBTF) and cadmium chloride (CdCl2) was determined with this method and the 50 % decreases in larval production are respectively 1,4 µg/l (TBTF) and 61 µg/l (Cd++).
